dCODE Dextramer® reagents can help identify new TCR sequences for subsequent expression of functional TCRs. In our latest poster from AACR, we showcase how we identified three α-β TCR sequences of interest from dCODE Dextramer® sequencing data on ligand-specific MAIT cells generated using the BD Rhapsody™ Single-Cell Analysis System. The TCR sequences were manufactured and refolded using our TCR Solutions platform, and evaluated for α-β pairing, functionality, and specificity using SDS PAGE and an in-house developed artificial cell (beads) assay. Two of the selected TCR sequences demonstrated correct folding, specificity, and functionality, as confirmed by their recognition of the 5-OP-RU MR1 Monomer on beads. Sepax Unix-C SEC columns were used in this recent Janssen paper to prepare and characterize the HIV-1 Env gp140, and then used in platform MASCALE (Mass Spectrometry Enabled Conversion to Absolute Levels of ELISA Antibodies). The HIV-1 Env gp140 proteins were produced in HEK293E cells and purified with Galanthus nivalis lectin bead-based affinity chromatography. Purified proteins show largely uniform trimeric content, with occasional hexamer contributions for some clade B strains.
